Protein S Functional Activity Test
Introduction to the Protein S Functional Activity Test
The Protein S Functional Activity Test is an essential diagnostic tool used to evaluate the functionality of Protein S, a protein that plays a critical role in the regulation of blood coagulation. This test is particularly important for individuals who may be at risk for thrombotic disorders, as it helps in identifying deficiencies that can lead to excessive clotting and other related complications.
What the Test Measures
This test specifically measures the functional activity of Protein S in the blood. Protein S works alongside Protein C to inhibit clot formation, thereby maintaining a balance in the coagulation process. A deficiency or dysfunction in Protein S can significantly increase the risk of developing blood clots.
Who Should Consider This Test
Individuals who may benefit from the Protein S Functional Activity Test include:
- Those with a personal or family history of thrombosis.
- Patients experiencing unexplained blood clots.
- Individuals with recurrent miscarriages or complications during pregnancy.
- Patients undergoing evaluation for clotting disorders.
Benefits of Taking the Test
Taking the Protein S Functional Activity Test provides several benefits, including:
- Early detection of coagulation disorders.
- Guidance for personalized treatment plans.
- Informed decision-making regarding lifestyle and medication changes.
- Peace of mind for patients and their families.
Understanding Your Results
Results from the Protein S Functional Activity Test will indicate whether your Protein S levels are within the normal range or if there is a deficiency. It is essential to discuss your results with a healthcare provider, who can provide insights into the implications and necessary follow-up actions.

- Transport to the lab within 4 hours. If this is not possible, make PPP within 1 hour of collection by centrifuging the sample at 3600 rpm for 15 minutes.
- Transfer the supernatant to a clean plastic tube and centrifuge again at 3600 rpm for 15 minutes.
- Transfer the final supernatant (PPP) to 1 labelled, clean plastic screw-capped vial and freeze immediately. Ship frozen. DO NOT THAW.
Overnight fasting is preferred. It is recommended to discontinue Heparin for 1 day and Oral Anticoagulants for 7 days prior to sampling, with prior consent from the treating physician. A duly filled Coagulation Requisition Form (Form 15) is mandatory.
