Sample Collection and Pre-Test Instructions
For the Factor X Functional Test, a sample of 3 mL whole blood is required in a Blue Top (Sodium Citrate) tube. It is crucial to mix the sample thoroughly by inversion and transport it to the lab within 4 hours. If immediate transport is not possible, it is recommended to prepare Platelet Poor Plasma (PPP) within 1 hour of collection. This involves centrifuging the sample at 3600 rpm for 15 minutes and transferring the supernatant to a clean plastic tube. The supernatant should then be centrifuged again and the final PPP transferred to a labelled, clean plastic screw-capped vial and frozen immediately.
Overnight fasting is preferred before the test, and a duly filled Coagulation Requisition Form (Form 15) is mandatory. It is also recommended to discontinue Heparin for 1 day and Oral Anticoagulants for 7 days prior to sampling, with prior consent from the treating physician.
