Protein S Antigen Total Test
Introduction
The Protein S Antigen Total Test is a vital diagnostic tool used to assess the levels of Protein S in the blood, a crucial protein that plays a key role in the regulation of coagulation. This test is important for identifying individuals with a deficiency of Protein S, which can lead to an increased risk of thrombosis (blood clots). The test is performed using an immunoturbidimetric method, ensuring accurate results for effective diagnosis and management.
What Does the Test Measure?
This test measures the total antigen levels of Protein S in the blood. Protein S is a vitamin K-dependent plasma protein that acts as a cofactor for activated protein C, helping to inactivate clotting factors Va and VIIIa, thus regulating blood clot formation.
Who Should Consider This Test?
This test is recommended for individuals who:
- Have a personal or family history of thrombotic events.
- Exhibit symptoms of thrombosis such as swelling, pain, or redness in limbs.
- Are being evaluated for unexplained blood clots.
- Have other risk factors for coagulation disorders.
Benefits of Taking the Test
Taking the Protein S Antigen Total Test offers several benefits:
- Helps in diagnosing Protein S deficiency, which is crucial for managing thrombotic risks.
- Guides treatment decisions for patients with coagulation disorders.
- Provides peace of mind for patients and their families regarding clotting risks.
Understanding Your Results
Results from this test will indicate the levels of Protein S in your blood. Normal levels suggest adequate Protein S function, while low levels may indicate a deficiency that could lead to an increased risk of clotting disorders. It is essential to discuss your results with your healthcare provider for appropriate management.

Sample Collection and Pre-Test Instructions
For this test, a sample of 3 mL whole blood is required in a 1 Blue Top (Sodium Citrate) tube. It is essential to mix the sample thoroughly by inversion and transport it to the lab within 4 hours. If this is not possible, make platelet-poor plasma (PPP) within 1 hour of collection by centrifuging the sample at 3600 rpm for 15 minutes and transferring the supernatant to a clean plastic tube. Centrifuge the supernatant again at 3600 rpm for 15 minutes and transfer the final supernatant (PPP) to a labeled clean plastic screw-capped vial. Freeze immediately and ship frozen. Do not thaw. Overnight fasting is preferred, and a duly filled Coagulation Requisition Form (Form 15) and Test Send Out Consent Form (Form 35) is mandatory.
